When to do a “single” collection vs “split”? It is an option on all the CCF forms!

DOT drug tests require a split specimen collection.
A minimum of 45 mL of urine is needed in total:

  • 30 mL is poured into Bottle A (for the initial test)
  • 15 mL is poured into Bottle B (for confirmatory or retest purposes, if requested)

This split ensures that a secondary test can be conducted if the donor disputes the original result, maintaining both accuracy and compliance with DOT regulations.


All Non-DOT drug tests are typically single-vial collections, unless otherwise specified by the requesting company.
While a single specimen vial is standard for Non-DOT testing, some companies may request a split specimen for added verification or internal policy reasons.

Always confirm the company’s testing requirements in advance to ensure proper collection and documentation.
